This study examined whether prior radiotherapy to the tumor site increases the efficacy of cemiplimab in patients with locally advanced or metastatic cutaneous SCC. Results suggested that previous radiation may enhance immunotherapy response, potentially through radiation-induced immunogenic cell death and neoantigen exposure.

Prior radiotherapy may enhance the efficacy of cemiplimab in advanced cSCC through synergistic immunogenic effects.

This radiation-immunotherapy interaction should be considered when sequencing treatments for advanced cutaneous SCC.
